Enclomiphene citrate binds to estrogen receptors in the hypothalamus and pituitary gland, acting as an estrogen antagonist in those tissues. By blocking estrogen’s action, it prevents the normal negative feedback that estrogen has on gonadotropin release. The brain essentially “senses” low estrogen, which triggers the hypothalamus to secrete GnRH and the pituitary to secrete more LH and FSH. Elevated LH then stimulates the Leydig cells in the testes to produce more testosterone, while FSH supports Sertoli cells and sperm production. Clinically, Enclomiphene has been shown to raise testosterone significantly in men with secondary hypogonadism. Unlike Clomiphene, which is a mix of isomers, Enclomiphene does not have the zuclomiphene component that can act like an estrogen agonist in some tissues. Therefore, Enclomiphene tends to avoid many side effects associated with Clomid (such as mood swings or blurred vision), while effectively restoring hormonal function.
